Regulatory role of l-proline in fetal pig growth and intestinal epithelial cell proliferation

l-proline (Pro) is a precursor of ornithine, which is converted into polyamines via ornithine decarboxylase (ODC). Polyamines plays a key role in the proliferation of intestinal epithelial cells. The study investigated the effect of Pro on polyamine metabolism and cell proliferation on porcine enterocytes in vivo and in vitro. Twenty-four Huanjiang mini-pigs were randomly assigned into 1 of 3 groups and fed a basal diet that contained 0.77% alanine (Ala, iso-nitrogenous control), 1% Pro or 1% Pro + 0.0167% α-difluoromethylornithine (DFMO) from d 15 to 70 of gestation. The fetal body weight and number of fetuses per litter were determined, and the small and large intestines were obtained on d 70 ± 1.78 of gestation. The in vitro study was performed in intestinal porcine epithelial (IPEC-J2) cells cultured in Dulbecco''s modified Eagle medium-high glucose (DMEM-H) containing 0 μmol/L Pro, 400 μmol/L Pro, or 400 μmol/L Pro + 10 mmol/L DFMO for 4 d. The results showed that maternal dietary supplementation with 1% Pro increased fetal weight; the protein and DNA concentrations of the fetal small intestine; and mRNA levels for potassium voltage-gated channel, shaker-related subfamily, member 1 (Kv1.1) in the fetal small and large intestines (P < 0.05). Supplementing Pro to either gilts or IPEC-J2 cells increased ODC protein abundances and polyamine concentrations in the fetal intestines and IPEC-J2 cells (P < 0.05). In comparison with the Pro group, the combined administration of Pro and DFMO reduced the expression of ODC protein and spermine concentration in the fetal intestine, as well as the concentrations of putrescine, spermidine and spermine in IPEC-J2 cells (P < 0.05). Meanwhile, the percentage of cells in the S-phase and the mRNA levels of proto-oncogenes c-fos and c-myc were increased in response to Pro supplementation, whereas depletion of cellular polyamines with DFMO increased tumor protein p53 (p53) mRNA levels (P < 0.05). Taken together, dietary supplementation with Pro improved fetal pig growth and intestinal epithelial cell proliferation via enhancing polyamine synthesis.     

汽车变速器齿轮修形仿真分析

汽车变速器的关键部件齿轮因其加工和装配质量的影响,工作时容易引起振动噪声大及承载能力降低等问题,而齿轮修形是解决这些问题的有效方法,且成本较低。针对某款汽车变速器,利用Romax对齿轮进行修形分析。结果表明,修形后减小了齿轮的传动误差和最大接触应力,齿面载荷分布更加合理。有效改善了齿轮的传动性能,增加了齿轮寿命。     

长江经济带国家级自然保护区内社区居民现状与发展对策

中国自然保护区多处于地理位置偏僻的贫困落后地区，且在有关法律条文规定下，居民对保护区内资源利用受限，特别是核心区和缓冲区中众多的社区居民给自然保护区管理带来压力，自然保护区保护与社区经济发展之间的矛盾日益凸显。文中通过对重点区域长江经济带国家级自然保护区内居民现状进行调查和分析，探讨不同类型自然保护区内社区居民人口密度、经济水平和自然资源依赖程度的差异，并针对自然保护区内社区发展存在的主要问题包括人口众多、经济水平受限等，提出降低保护区内人口密度、多形式提高居民收入、发展生态旅游和加强可持续经营管理等建议。     

空气源热泵烤房与生物质新能源烤房对烤后烟叶经济效益的影响

为空气源热泵烤房及生物质新能源烤房的推广提供参考，比较常规燃煤烤房、空气源热泵烤房及生物质新能源烤房烘烤烟叶的用工成本、能耗成本、烟叶等级及经济性状差异。结果表明：空气源热泵烤房烤后烟叶经济性状最佳，上等烟率、中等烟率、桔黄烟率分别达81.00%、19.00%和95.05%，下等烟率为0.00%；用工成本、能耗成本空气源热泵烤房均比生物质新能源烤房和常规燃煤烤房低40%以上，综合效益提高13%以上，差异显著。     

上海市规模化奶牛场布鲁氏菌病和牛结核病风险评估分级体系研究

为尽快实现上海崇明奶牛布鲁氏菌病和牛结核病(以下简称"两病")区域净化示范区净化目标,通过建立奶牛场"两病"输入性风险评估和内部风险评估模型,结合奶牛场"两病"流行史,构建了上海市规模化奶牛场"两病"风险评估分级体系;应用该体系将奶牛场划分为高、中、低风险场,为实施分类管理策略奠定基础。运用该体系,对上海市崇明区奶牛场每年开展1次"两病"风险评估分级。经过连续3年的风险分级与管理,崇明区奶牛场的"两病"净化效率得到极大提高,并使崇明奶牛"两病"区域净化示范区顺利通过国家验收,表明该体系符合上海市奶牛"两病"净化工作实际。

利用重组自交系高密度遗传图谱定位水稻芽期耐冷QTL

为定位水稻芽期耐冷QTL,本实验以双季超级稻品种‘五丰优T025’的双亲‘五丰B’和‘昌恢T025’杂交衍生的重组自交系(recombinant inbred lines, RILs)群体为材料,对10℃低温处理的水稻幼芽的存活率、根数、根长和芽长进行了测定。利用QTL Icimapping v4.2软件,共检测到3个控制芽期耐冷性QTL：qRL1、qRL2和qBL6,分别位于第1、2、6染色体上,LOD值分别为2.98,2.51和5.26,分别解释表型变异的10.54%,8.67%和14.04%,其增效等位基因均来自于亲本‘昌恢T025’。这些QTL定位在6.75k~40.05 kb染色体区间,为后续利用这些QTL进行分子标记辅助,选育芽期耐冷籼稻新品种奠定了基础。此外,检测到13对影响水稻芽期耐冷上位性互作QTL,分布在所有12条染色体,其中第3染色体与第8染色体之间互作位点可解释的表型变异率达到21.77%,表明上位性互作QTL在调控水稻芽期耐冷过程中也发挥了重要作用。     

Effects of glycinin on growth performance,immunity and antioxidant capacity in juvenile golden crucian carp,Cyprinus carpio × Carassius auratus

This study was designed to examine the effects of glycinin on growth, digestive ability, immune responses, antioxidant capacity and gene expression levels of golden crucian carp. Golden crucian carp were fed diets containing glycinin at 0, 30, 60, 90 and 120 g/kg, respectively, for 8 weeks. Body weight, weight gain percentage, specific growth rate and feed efficiency ratio were negatively related to the content of glycinin in diet. Activities of protease, acid phosphatase, alkaline phosphatase, lysozyme in hepatopancreas, and activities of catalase, glutathione peroxidase, superoxide dismutase, and total antioxidant capacity in the proximal intestine, mid intestine, distal intestine and hepatopancreas were negatively related to the content of glycinin in diet, whereas malondialdehyde in proximal intestine, mid intestine, distal intestine and hepatopancreas increased directly with the content of glycinin in diet. Furthermore, the relative expressions of TNF‐α and IL‐1β in proximal intestine, mid intestine and distal intestine increased directly with the content of glycinin in diet, whereas the relative expressions of TNF‐α and IL‐1β in hepatopancreas were negatively related to the content of glycinin in diets. The relative expressions of IL‐10 in proximal intestine, mid intestine, distal intestine and hepatopancreas all were negatively related to the content of glycinin in diets. In conclusion, reductions in growth, immunity and antioxidant capacity, intestine inflammation with dysfunction of digestive system occurred in golden crucian carp that fed a diet containing glycinin at 30 g/kg or higher after 8 weeks.     

出口动物及动物产品传带新型冠状病毒风险评估

在全球多地发生新型冠状病毒肺炎(COVID-19)疫情的背景下,已有部分国家(地区)对我国货物贸易进行管制。为维护我国正常出口贸易秩序,参考世界卫生组织、世界动物卫生组织等国际组织的疫病风险分析流程,对我国出口动物及动物产品传带新型冠状病毒(SARS-CoV-2)的风险进行定性分析。分析认为:目前我国出口活动物感染或携带SARS-CoV-2的风险极低;按照正常程序生产和加工的出口动物源性产品携带、污染并传播SARS-CoV-2的风险极低;我国相关部门已采取了的严格的防控措施,最大程度降低了出口动物及动物源性产品的SARS-CoV-2污染风险。综上,我国出口动物及动物产品携带或传播SARS-CoV-2的风险极低。建议各国通过共同协作,加强COVID-19防控,以降低COVID-19对相互间出口贸易的影响。
